Beschrijving
- Recognizes human FN1
- Fibronectin 1 (FN1) is a high molecular weight glycoprotein, existing in soluble form in plasma (plasma FN1) and other body fluids and in insoluble form in extracellular matrix (cellular FN1)
- Plasma FN1 (dimeric form) secreted by hepatocytes
- Cellular FN (dimeric or cross-linked multimeric forms) made by fibroblasts, epitherlia and other cell types, deposited as fibrils in extracellular matrix
- FN1 binds to cell surfaces through integrins and components (collagen, fibrin, heparin)
- Involved in cell adhesion and migration processes (embryogenesis, wound healing, hemostasis, host defense, metastasis)
Process:
- Antibody specific for FN1 pre-coated onto microwells
- FN1 protein in samples captured by the coated antibody after incubation
- After extensive washing, another antibody specific for FN1 is added to detect captured FN1 protein
- For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
