- Recognizes human angiogenin
- Angiogenin (ANG), an angiogenic ribonuclease, is a member of vertebrate-specific secreted RNase superfamily
- Identified as tumor angiogenic factor, related to growth and metastasis of tumors
- Proposed as permissive factor for angiogenesis induced by other angiogenic factors (VEGF, basic fibroblast growth factor, acidic fibroblast growth factor, epidermal growth factor)
- Increased angiogenin serum levels associated with incidence and severity of human tumors including HCC
Process:
- Antibody specific for angiogenin pre-coated onto microwells
- Angiogenin protein in samples captured by the coated antibody after incubation
- After extensive washing, another antibody of biotinylated specific for angiogenin is added to detect captured angiogenin protein
- For signal development, Sterptravidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
